CJC-1295 with or without DAC: the difference

Both forms share the same base: the first 29 amino acids of human GHRH. DAC is not a dosage — it is a chemical group. The Drug Affinity Complex is a maleimide grafted onto the peptide that binds covalently to serum albumin, protecting it from rapid clearance.

The result is a difference in kinetics, not potency: a half-life described at 5.8 to 8.1 days for the DAC form (Teichman et al., 2006) versus a few tens of minutes without DAC. This reference is the no-DAC version, chosen when a study protocol aims to reproduce a short, near-physiological pulse rather than sustained exposure.

Why pair a GHRH analogue with a secretagogue

CJC-1295 binds the GHRH receptor (Gs-coupled, raising intracellular cAMP); Ipamorelin acts on the growth-hormone secretagogue receptor (GHS-R, Gq-coupled, mobilising the phospholipase C / calcium pathway). The pathways are distinct, and GHRP work also describes attenuation of somatostatin — the physiological brake on GH. In models, combining the two yields a response greater than the sum of either stimulus alone.

Ipamorelin is preferred over older GHRPs for its selectivity: Raun et al. (1998) reported no notable rise in ACTH, cortisol or prolactin at doses well above those producing GH release in animals, unlike GHRP-6.

Reconstitution: a worked example

With 2 mL of bacteriostatic water in a 5 mg + 5 mg vial: 5 mg ÷ 2 mL = 2.5 mg/mL, i.e. 2,500 mcg/mL of each compound. On a U-100 syringe, 10 IU = 0.1 mL = 250 mcg of each compound, giving 20 draws of 10 IU per vial.

The most common trap: the volume of water never changes how much peptide is in the vial. Reconstituting with 1 mL instead of 2 mL gives 5 mg/mL instead of 2.5 mg/mL — the same total mass in half the volume. Only the volume to draw changes.
